Figure 1. Jurkat cells, grown in suspension, were incubated with 1 µM staurosporine for 3 hours at 37°C to induce apoptosis, and then labeled with SR-LEHD-FMK (Cat. 960/961) for 60 minutes at 37°C. Slides were prepared and imaged using a fluorescence microscope with a broad band pass filter. Figure 2. Jurkat cells were exposed to two experimental conditions (A and B). Each condition was treated with either DMSO (negative) or staurosporine (apoptotic), then labeled with SR-LEHD-FMK (Cat. 960/961). Samples were read in a fluorescence plate reader (550 nm excitation and 590 nm emission using a 570 nm cut-off filter). Figure 2. Paramecium were starved for 12-24 hours in 5 mM KCl to induce autogamy. SR-VAD-FMK (Cat. 916/917), was added to the live Paramecium for 30 minutes in the dark. Cells were then washed and Hoechst 33342 was added. Cells were incubated in the dark for 10 minutes, washed, and applied to coverslips. Figure 1. Jurkat cells were treated with staurosporine (Positive, Induced, right histogram). Negative control cells were spiked with a DMSO vehicle control (Negative, Non-induced, left histogram). Cells were stained with SR-VAD-OPH for 1 hour, washed twice, and read on a flow cytometer. Only 8.0% of control cells displayed caspase activity, whereas staurosporine induced caspase activity in 90.3% of the experimental cells. This is a ratio of 11:1. Figure 2. Jurkat cells were treated with staurosporine to induce apoptosis, then labeled with SR-VAD-OPH for 60 minutes. Cells were washed twice and wet-mount slides were prepared. A grey DIC image (left) was taken, and red fluorescence (right) was detected using a band pass filter (excitation at 565 nm, emission at 590-600 nm).
