Methotrexate (MTX) is a cytotoxic agent known to inhibit dihydrofolate reductase, a crucial enzyme in folic acid metabolism. It is actively taken up by cells and binds to dihydrofolate reductase, thereby inhibiting folic acid metabolism and inducing cell cycle arrest. Methotrexate conjugates, which combine the chemotherapeutic agent methotrexate with fluorescent dyes, are most commonly used in targeting cancer cells, to study how drugs bind to their target receptors, antimetabolite resistance, and spontaneous gene amplification. These processes are all involved in cellular resistance to certain drugs, thus aiming to improve drug delivery and efficacy.
