Anti-TMEM240 Recombinant Chicken Chimeric mAb (N498/28)
Our chicken Anti-TMEM240 recombinant monoclonal antibody is a chimeric antibody derived from NeuroMab clone N498/28. It detects human, mouse, and rat TMEM240 and is purified by affinity chromatography. It works in ELISA, ICC, IHC, WB.
Immunohistochemistry for detection of TMEM240 in adult rat hippocampus using Aves Labs’ anti-TMEM240 recombinant chicken monoclonal antibody [N498/28] (Cat No. 78-533). Tissue was fixed with 4% paraformaldehyde and paraffin embedded. Heat induced antigen retrieval was performed using citrate buffer (Cat No. AR-6544) and then blocked with Immuno Protein Blocking Solution with Goat Serum (Cat No. AR-6591). The anti-TMEM240 antibody (Cat No. 78-533) was incubated with the sample at a dilution of 1:250 at 4C overnight. After washing with PBST, the sections were stained using goat anti-chicken IgY CF488 secondary antibody (Cat No. 80-1001-488) (green). DAPI nuclear stain (blue) shows cell nuclei.

TMEM240
The TMEM240 (Transmembrane Protein 240) gained significant attention in the 2010s when it was linked to the neurodegenerative disorder Spinocerebellar Ataxia Type 21 (SCA21). TMEM240 is a small transmembrane protein highly expressed in the central nervous system, particularly within the Purkinje cells of the cerebellum and the synapses of the cerebral cortex. While its precise biochemical pathway is still being studied, research suggests it plays a vital role in maintaining synaptic integrity and intracellular signaling. Mutations in the TMEM240 gene lead to the misfolding or loss of function of the protein, which triggers the progressive motor incoordination and cognitive impairment characteristic of SCA21.

As such this antibody retains the same binding performance as the original clone N498/28 but can be detected using standard anti-chicken secondary antibodies allowing flexibility for multiplexing applications. This antibody is expressed recombinantly in mammalian cells and then affinity purified from the cell culture media.
