Scientific Background

The Ryanodine Receptor 2 (RyR2) is important in cardiac physiology, historically identified through its high affinity for the plant alkaloid ryanodine. First cloned and characterized in the late 1980s, it is key in understanding of how the heart beats at a molecular level. Unlike its skeletal muscle counterpart (RyR1), RyR2 is the primary calcium release channel located on the sarcoplasmic reticulum (SR) of cardiac muscle cells. Biologically, RyR2 is the gatekeeper of excitation-contraction coupling. When an action potential triggers a small influx of calcium through L-type channels, RyR2 senses this and responds by releasing a large "spark" of calcium from internal stores into the cytosol—a process known as calcium-induced calcium release (CICR). This sudden surge in Ca2+ concentration allows the heart muscle to contract. Because of its large size and complex regulatory domains, RyR2 is a hotspot for genetic mutations; even slight "leaks" in the channel during the heart's resting phase can lead to fatal arrhythmias, such as Catecholaminergic Polymorphic Ventricular Tachycardia (CPVT), or contribute to chronic heart failure.

Production Notes

This recombinant antibody is a chimeric antibody created by replacing the mouse heavy and light constant regions of clone N501/49 with chicken IgY heavy and light constant regions. As such this antibody retains the same binding performance as the original clone N501/49 but can be detected using standard anti-chicken secondary antibodies allowing flexibility for multiplexing applications. This antibody is expressed recombinantly in mammalian cells and then affinity purified from the cell culture media and buffer exchanged into 1X PBS.
