Product Details
Rab 9
Rab9 is a small GTP-binding protein of the Rab family that regulates late endosomal trafficking. It is primarily involved in the transport of mannose-6-phosphate receptor-containing vesicles from late endosomes back to the trans-Golgi network, a pathway critical for proper lysosomal enzyme sorting and recycling. Like other Rab proteins, Rab9 functions as a molecular switch, cycling between an active GTP-bound state and an inactive GDP-bound state, with its activity controlled by guanine nucleotide exchange factors (GEFs) and GTPase-activating proteins (GAPs). Rab9-mediated trafficking ensures the steady-state distribution of cation-dependent and cation-independent mannose-6-phosphate receptors and contributes to lysosomal homeostasis. Dysregulation of Rab9 function has been linked to impaired lysosomal degradation, altered receptor recycling, and has emerging associations with cancer progression, viral infection, and lysosomal storage disorders.

This recombinant antibody is a chimeric antibody created by replacing the mouse heavy and light constant regions of clone Rab9, clone 9 with chicken IgY heavy and light constant regions. As such this antibody retains the same binding performance as the original clone Rab9, clone 9 but can be detected using standard anti-chicken secondary antibodies allowing flexibility for multiplexing applications. This antibody is expressed recombinantly in mammalian cells and then affinity purified from the cell culture media.

UniProt Summary: The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes. Rabs cycle between an inactive GDP-bound form and an active GTP-bound form that is able to recruit to membranes different sets of downstream effectors directly responsible for vesicle formation, movement, tethering and fusion. RAB9A is involved in the transport of proteins between the endosomes and the trans-Golgi network (TGN). Specifically uses NDE1/NDEL1 as an effector to interact with the dynein motor complex in order to control retrograde trafficking of RAB9-associated late endosomes to the TGN. Involved in the recruitment of SGSM2 to melanosomes and is required for the proper trafficking of melanogenic enzymes TYR, TYRP1 and DCT/TYRP2 to melanosomes in melanocytes.
