Anti-PSD-95 Recombinant Guinea Pig Chimeric mAb (K28/43)
Our guinea pig Anti-PSD 95 recombinant monoclonal antibody is a chimeric antibody derived from NeuroMab clone K28/43. It detects chicken, human, mouse, non-human primate, and rat PSD 95 and is purified by affinity chromatography. KO validated. Works in WB, IHC, ICC, IP, ELISA.
Immunohistochemistry for detection of PSD-95 in adult rat cerebellum using Anti-PSD-95 Recombinant Chimeric Guinea Pig Monoclonal antibody [K28/43] (Cat No. 74-028). Tissue was fixed with 4% paraformaldehyde and paraffin embedded. Heat induced antigen retrieval was performed using citrate buffer (Cat No. AR-6544) and then blocked with Immuno Protein Blocking Solution with Goat Serum (Cat No. AR-6591). The anti-PSD-95 antibody (Cat No. 74-028) was incubated with the sample at a dilution of 1:250 at 4C overnight. After washing with PBST, the sections were stained using goat anti-guinea pig 488 secondary antibody (green). DAPI nuclear stain (blue) shows cell nuclei. Anti-PSD-95 specifically stains the excitatory synapses in the granular layer of the cerebellum.

UniProt Summary: Postsynaptic scaffolding protein that plays a critical role in synaptogenesis and synaptic plasticity by providing a platform for the postsynaptic clustering of crucial synaptic proteins. Interacts with the cytoplasmic tail of NMDA receptor subunits and shaker-type potassium channels. Required for synaptic plasticity associated with NMDA receptor signaling. Overexpression or depletion of DLG4 changes the ratio of excitatory to inhibitory synapses in hippocampal neurons. May reduce the amplitude of ASIC3 acid-evoked currents by retaining the channel intracellularly. May regulate the intracellular trafficking of ADR1B. Also regulates AMPA-type glutamate receptor (AMPAR) immobilization at postsynaptic density keeping the channels in an activated state in the presence of glutamate and preventing synaptic depression. Under basal conditions, cooperates with FYN to stabilize palmitoyltransferase ZDHHC5 at the synaptic membrane through FYN-mediated phosphorylation of ZDHHC5 and its subsequent inhibition of association with endocytic proteins.
