Anti-ZsGreen Antibody

Our Anti-ZsGreen chicken polyclonal primary antibody from Aves Labs is produced in-house. It detects ZsGreen, ZsGreen1, ZsGreen1-DR and other fluorescent proteins derived from ZsGreen. It is affinity-purified IgY and is great for use in ICC, IHC, WB.

Immunofluorescent staining of ZsGreen-transfected COS-7 cells using Aves Labs chicken polyclonal anti-ZsGreen antibody (red, Cat No. ZSGREEN-0100) and showing ZsGreen autofluorescence (green). The cells were fixed with 3% paraformaldehyde, washed with PBS, and blocked/permeabilized with 5% non-fat milk powder in PBST. The anti-ZsGreen chicken polyclonal antibody (Cat No. ZSGREEN-0100) was incubated with the sample at a dilution of 1:500 (0.4μg/mL) or 1:1000 (0.2 μg/mL) at 4C overnight. After washing with PBST, the cells were stained using goat anti-chicken IgY CF594 secondary antibody (Cat No. 80-1001-594) (red). The staining revealed a complete overlap between the signal from the chicken anti-ZsGreen antibody and the ZsGreen autofluorescence in transfected cells. The cells were mounted with Antibodies Incorporated Fluoroshield with DAPI mounting medium (Cat No. AR-6501). DAPI nuclear stain (blue) shows cell nuclei.
Western blot detection of ZsGreen in mock-transfected (lane 1) and ZsGreen-transfected (lane 2) COS-7 cell lysates using Aves Labs anti-ZsGreen chicken polyclonal antibody (Cat No. ZSGREEN-0100). 10-20% Tris-Glycine gel was loaded with 10ug of mock-transfected and ZsGreen-transfected COS-7 cell lysates. Proteins were transferred to PVDF membrane and blocked with 5% non-fat milk powder. Anti-ZsGreen antibody was incubated with the membrane at a concentration of 0.08 µg/ml (1:2500) and then detected via chemiluminescence (30 sec) using a Goat anti-Chicken H+L, HRP conjugated secondary antibody. Anti-ZsGreen recognizes exogenous ZsGreen only in the ZsGreen-transfected cell lysates at the expected molecular weight of ~26 kDa. Detection of endogenous Calnexin using Aves Labs anti-Calnexin chicken polyclonal antibody (Cat No. CANX-1000) was used as a loading control.
