Anti-Synuclein, pan Antibody

Our Anti-Pan-synuclein rabbit polyclonal primary antibody detects human, mouse, and rat Pan-synuclein, and is affinity-purified. It is validated for use in FC, WB.

Detection of synuclein expression in tissue homogenate and cell lysates by Western Blot. Synuclein monomer is detected at ~14-18 kDa, additional higher-molecular weight bands may represent multimeric forms of synuclein. Method: Reducing SDS-PAGE; Transfer: Towbin’s buffer with 20% methanol; Protein fixation: proteins were fixed on membrane (0.4% formaldehyde, 30 minutes at room temperature) after transfer, as described by Lee & Kamitani, 201; Blocking: 5% skim milk, 1 hour at room temperature; Primary antibody: 1.5 μg/mL, overnight incubation at 2-8°C; Secondary antibody: anti-sheep-HRP (1:6,000); Detection: Chemiluminiscence. Peptide blocking was carried out in presence of two-fold mass excess of peptide over antibody.
Analysis of pan-synuclein expression in rat PC-12 and C6 cell lines by Flow Cytometry. Cells were fixed and permeabilized in methanol (10 minutes). Blocking: 15% horse serum, 30 minutes on ice; Primary antibody: Rabbit polyclonal antibody to pan-synuclein (R-2110-100, red, 20 µg/mL), 60 minutes on ice. Secondary antibody: donkey anti-rabbit-PE (1:100 dilution), 30 minutes in dark on ice. Negative control: Non-specific Control rabbit IgG, (black dashed). Data and results were generated using Orflo MoxiGoTM instrument and protocols.
