Anti-GluN2A/NR2A Glutamate Receptor Recombinant Chicken Chimeric mAb (N327/95)
Our chicken Anti-GluN2A/NR2A glutamate receptor recombinant monoclonal antibody is a chimeric antibody derived from NeuroMab clone N327/95. It detects human, mouse, rabbit, and rat GluN2A/NR2A glutamate receptor and is purified by affinity chromatography. Works in WB, IHC, ICC, ELISA.
Representative section of formalin fixed, paraffin-embedded rat brain cerebellum showing cells stained by anti-GluN2A/NR2A Glutamate Receptor recombinant chicken mAb (green, Cat No. 78-288) using goat anti-chicken IgY CF®488 secondary antibody (Cat No. 80-1001-488). The stained sections were mounted with Antibodies Incorporated’s Fluoroshield with DAPI and DABCO mounting medium (Cat No. AR-6505). DAPI nuclear stain (blue) shows cell nuclei.

Product Details
GluN2A/NR2A glutamate receptor
Glutamate receptor NMDA2A or GluN2A/NR2A glutamate receptor is a member of the glutamate-gated ion channel family. NR2A is a subunit of the ligand-gated ion channel with high calcium permeability and voltage-dependent sensitivity to magnesium. NR2A is expressed throughout the brain, including striatum, forbrain, cerebellum (at protein level) brain cortex, piriform cortex, hippocampus, caudate-putamen, dentate gyrus and granule cell layer. It is found at the postsynaptic cell membrane, dendritic spine, synapse, and cytoplasmic vesicle membrane. A variety of neurological diseases, including epilepsy, intellectual disability, autism spectrum disorders, developmental delay, and schizophrenia are associated with mutations in the NR2A gene.

This recombinant antibody is a chimeric antibody created by replacing the mouse heavy and light constant regions of clone N327/95 with chicken IgY heavy and light constant regions. As such this antibody retains the same binding performance as the original clone N327/95 but can be detected using standard anti-chicken secondary antibodies allowing flexibility for multiplexing applications. This antibody is expressed recombinantly in mammalian cells and then affinity purified from the cell culture media.

UniProt Summary: Component of N-methyl-D-aspartate (NMDA) receptors (NMDARs) that function as heterotetrameric, ligand-gated cation channels with high calcium permeability and voltage-dependent block by Mg(2+). NMDARs participate in synaptic plasticity for learning and memory formation by contributing to the slow phase of excitatory postsynaptic current, long-term synaptic potentiation, and learning. Channel activation requires binding of the neurotransmitter L-glutamate to the GluN2 subunit, glycine or D-serine binding to the GluN1 subunit, plus membrane depolarization to eliminate channel inhibition by Mg(2+). NMDARs mediate simultaneously the potassium efflux and the influx of calcium and sodium. Each GluN2 subunit confers differential attributes to channel properties, including activation, deactivation and desensitization kinetics, pH sensitivity, Ca2(+) permeability, and binding to allosteric modulators. Participates in the synaptic plasticity regulation through activation by the L-glutamate releaseed by BEST1, into the synaptic cleft, upon F2R/PAR-1 activation in astrocyte.
