Anti-Phospho-GUK1 (Tyr74) Antibody
Our GUK1 (Tyr74) rabbit polyclonal phosphospecific primary antibody from PhosphoSolutions is produced in-house. It detects human, mouse, and rat GUK1 (Tyr74) and is antigen affinity purified from pooled serum. It is great for use in WB, IHC, ICC, ELISA.
Western blot detection of GUK1 phosphorylated at Tyr74 using anti-pGUK1 Tyr74 rabbit polyclonal antibody (Cat No. pGUK1-74 ). Whole cell lysates were generated from HEK293T cells stably co-expressing EML4-ALK fusion protein with FLAG-GUK1-WT or FLAG-GUK1-Y74F mutant, run on SDS-PAGE gel, transferred to a nitrocellulose membrane and examined by Western blot. The blot was probed with pGUK1-Y74 primary antibody at a concentration of 1:200 and then detected via chemiluminescence using a Goat Anti-Rabbit H+L, HRP conjugated secondary antibody. Actin was used as a loading control to show equal loading of both lysates. pGUK1 Y74 antibody recognizes only GUK1-WT and not the phospho-deficient mutant GUK1-Y74F at the expected molecular weight of ~25 kDa. GUK1 (Tyr74)
Guanylate kinase 1 (GUK1) is an essential enzyme in the purine salvage pathway, catalyzing the reversible conversion of GMP to GDP. This reaction is a rate-limiting step in maintaining the cellular GTP pool required for nucleic acid synthesis, signal transduction, and energy metabolism. Clinically, GUK1 is vital for the metabolic activation of guanine-based prodrugs, making it a determinant of antiviral and chemotherapeutic efficacy. GUK1 phosphorylation at Tyr74 by non-receptor tyrosine kinases (e.g., Src or Syk) has been linked to increased nucleotide production in hyper-proliferative states. Elevated levels of Tyr74-phosphorylated GUK1 are frequently observed in various malignancies, where they support the metabolic demands of rapid tumor growth, positioning this specific phospho-form as a significant biomarker for metabolic reprogramming in oncology and drug resistance studies. It is phosphorylated by the fusion kinase EML4-ALK in lung cancers, creating a metabolic dependency, and making it a potential target for therapy.
