Hybridoma mAb Scale-Up

Once a mouse monoclonal cell line has been sub-cloned to monoclonality, it can be used as an infinite source of monoclonal antibody (mAb) through either in vitro (bioreactor) or in vivo (ascites) production.

Method Overview: In Vitro Bioreactor Production

Cell-line Expansion (1-2 weeks)

  • Monoclonal cell line is thawed and expanded for seeding bioreactor.
  • Cell-line adaptation to serum-free medium available upon request (adaptation will take up to 3 additional weeks).

Bioreactor Production (2 weeks)

  • Cell line is seeded into bioreactor and antibody produced.

mAb Purification

 

Method Overview: In Vivo Ascites Production

Cell-line Expansion (Project-dependent timeline)

  • Monoclonal cell line is thawed and expanded for seeding mice.

Inoculation (3-4 weeks)

  • Mice are primed, cells seeded, then antibody harvested.

mAb Purification (3 weeks)
